Tight junctions between epithelial and endothelial cells form selective barriers and paracellular channels that regulate paracellular transport of solutes, immune cells, and drugs. One goal of current tight junction research is to clarify the relationship between the molecular basis of the junctions and physiological functions and pathologies. This volume presents a collection of contributions by leading scientists working in the field on topics such as functions of distinct tight junction proteins (occludin, tricellulin, claudin) as barrier or channel-formers for solutes and water; characteristics of the tight junction in inflammatory bowel disease; posttranslational modifications of tight junction proteins; the relationship between renal tight junction proteins and blood pressure control; and the molecular structure of claudin-claudin interactions. The results presented here may form a basis for future diagnostic and therapeutic approaches to diseases that may not seem to have much in common but are in fact characterized by defects of organ barriers, diseases such as Crohn's disease, renal hypertension, inner ear deafness, and cancer.
